Reproducibility of MDM2 and CDK4 staining in soft tissue tumors.

چکیده
MDM2 and CDK4 immunostaining can be useful adjuncts in diagnosing liposarcoma among soft tissue neoplasms. We examined the reproducibility of MDM2 and CDK4 staining between 2 laboratories and between tissue microarrays and whole tissue sections. Sixty-two soft tissue tumors were immunostained at the Bergonié Institute, Bordeaux, France, and the Curie Institute, Paris, France. We also examined 203 soft tissue neoplasms on standard tissue sections and tissue microarrays. There was high concordance of results obtained from the 2 laboratories (with 2 different pathologists) for MDM2 (kappa, 0.93) and CDK4 (kappa, 0.8) staining. There also was excellent concordance between results on tissue microarray and on whole tissue sections for MDM2 (kappa, 0.80) and CDK4 (kappa, 0.93). Immunostaining for MDM2 and CDK4 is a reproducible technique that may be exported to different laboratories for routine use. Tissue microarray is indicated for studying large series.

Mdm2 and p53 expressions and Ki-67 proliferative index in fibrohistiocytic tumors
Objective: Various genetic alterations including inactivation of tumor suppressor genes and activation of oncogenes, are described in different types of neoplasms as well as soft-tissue tumors. The purpose of the present study is to evaluate the expressions of mdm2 and p53, and Ki-67 proliferative activity and to investigate their interrelations with the clinicopathologic parameters in fibrohis...
متن کاملMDM2 and CDK4 amplifications are rare events in salivary duct carcinomas
Salivary duct carcinoma (SDC) is an aggressive adenocarcinoma of the salivary glands associated with poor clinical outcome. SDCs are known to carry TP53 mutations in about 50%, however, only little is known about alternative pathogenic mechanisms within the p53 regulatory network. Particularly, data on alterations of the oncogenes MDM2 and CDK4 located in the chromosomal region 12q13-15 are lim...
متن کاملDifferential diagnosis of atypical lipomatous tumor/well-differentiated liposarcoma and dedifferentiated liposarcoma: utility of p16 in combination with MDM2 and CDK4 immunohistochemistry.
The differential diagnosis between atypical lipomatous tumor/well-differentiated liposarcoma (ALT/WDLPS) and dedifferentiated liposarcoma (DDLPS) from their morphologic counterparts is challenging. Currently, the diagnosis is guided by MDM2 and CDK4 immunohistochemistry (IHC) and is confirmed by the amplification of the corresponding genes. Recently, p16 IHC has been proposed as a useful diagno...
متن کاملRelationship between grade and MDM2 oncoprotein overexpression in transitional cell carcinoma of the urinary bladder
Objective: Transitional cell carcinoma (TCC) of the urinary bladder is the second common cancer of the genitourinary tract. Several parameters such as clinical and pathological parameters, molecular factors, and etc play a role in determination of prognosis and type of treatment. In this research study, ...
متن کاملAmplification of multiple genes from chromosomal region 12q13-14 in human malignant gliomas: preliminary mapping of the amplicons shows preferential involvement of CDK4, SAS, and MDM2.
We have investigated 234 tumors of the central nervous system for amplification of 9 different loci from 12q13-14 and report that about 15% of the anaplastic astrocytomas and glioblastomas show amplification at this chromosomal region. The genes most frequently amplified were CDK4 and SAS (18 of 19 cases). MDM2 was coamplified with CDK4 and SAS in 11 tumors while one glioblastoma showed only MD...
